摘  要:目的研究端粒酶阴性不依赖端粒酶端粒维持旁路(ALT)细胞与端粒酶阳性肿瘤细胞侵袭能力差异。方法应用Transwell法检测7种端粒酶阴性ALT细胞系和7种端粒酶阳性细胞系的侵袭能力,应用Oligo基因芯片技术和实时定量RT-PCR分析两类细胞的金属基质蛋白酶家族的表达差异。结果端粒酶阴性ALT细胞系的侵袭能力显著低于端粒酶阳性肿瘤细胞。端粒酶阴性ALT细胞系的MMP-1、MMP-7的表达显著低于端粒酶阳性肿瘤细胞,而TIMP-1、-2、-3显著高于端粒酶阳性肿瘤细胞。结论端粒酶阴性ALT细胞系的侵袭能力显著低于端粒酶阳性肿瘤细胞,这是端粒酶阴性ALT肿瘤预后好于端粒酶阳性肿瘤的重要原因之一,MMP-1和MMP-7的低表达以及TIMP-1、-2、-3的高表达是其侵袭能力低的重要机制。Objective To study the difference of invasion ability between multiple ALT cell lines and multiple telomerase positive cancer cell lines. Method Transwell plat coated matrigel model was used to investigate the invading ability of 7 ALT cell lines and 7 telomerase positive cell lines. Oligonucleotide microarray gene chip analysis and real time PCR were used to screen and confirm the expression difference of matrix metalloproteinase family in multiple ALT cell lines comparing with multiple telomerase positive cancer cell lines. Results Seven hundred different genes were detected, the different genes included the genes involving the matrix degrading. TIMP expression in the ALT cell lines was higher than telomerase positive cell lines, and the MMP-1 and MMP-7 expression were lower. Conclusion The invading ability of ALT cell lines was significantly weaker than that of telomerase positive cell lines, which was one of the key mechanism of the better prognosis of ALT tumors. The low expression of MMP-7 and MMP-1 and the high expression of TIMP-1 ,-2 ,-3 were one of the mechanisms of its weak invading ability.
