摘 要:目的:选择一种新的交联剂制备栓塞用平阳霉素明胶微球。方法:分别以氧化葡聚糖和戊二醛作为交联剂制备2种注射用盐酸平阳霉素(PYM)明胶微球,比较2种微球形态学及生物学特性。结果:光镜下葡聚糖组微球的圆整度均优于戊二醛组;图像分析结果显示2组微球的粒径均符合栓塞用微球的要求;与戊二醛组相比,氧化葡聚糖组的粒径分布范围较窄(P<0.05);包封率、载药率(94.6%、8.9%)均高于戊二醛组(80.1%、5.5%),具有显著差异(P<0.05);且6h的累积释放率(71%)低于戊二醛组(91%),释药缓慢。结论:氧化葡聚糖作为一种新的交联剂具有更突出的优点。Objective:To choose a new crosslinking agent to prepare gelatin-Pingyangmycin(PYM)-microspheres for the interventional chemoemoblization therapy, nethods:Oxidized-dextran and glutaraldehyde were used respectively as crosslinking agent to prepare two kinds of gelatin-PYM-microspheres. The characteristics of prepared microspheres with different crosslinking agents were compared. Results:The roundness of oxidized-dextran group is better than glutaraldehyde group through optical microscope observation; and the distribution range of particle diameter of the two kinds of microspheres were fit for the use of embolization. However, oxidized-dextran group was with a narrower size distribution when compared with glutaraldehyde group( P 〈 0.05 ) ; the drug content rate and the encapsulation rate of oxidized-dextran group were 94.6%, 8.9% respectively, which were higher than glutaraldehyde group, whose data were 80.1% , 5.5% ( P 〈0.05 ) ; the cumulative release rate of PYM from oxidized-dextran group in 6 hours was 71%, which was lower than glutaraldehyde group 95% ( P 〈 0.05 ). Conclusion: These results implied the oxidized-dextran as a new kind of crosslinking agent has more advantages than glutaraldehyde.
