摘  要:目的通过观察Ibrolipi m对贵州小型猪(小型猪)的胰岛结构和功能的影响,探讨其调节糖代谢的作用机制。方法小型猪15头,随机分为3组。分别喂普通猪饲料、高脂高糖饲料(HFSD)和HFSD加1%Ibrolipi m。每月末测定血糖(PG)和胰岛素(Ins)浓度。第6个月末作OGTT和胰岛素敏感性试验。第8个月末处死动物,取胰腺标本,作胰岛素含量测定。用HE染色、免疫组化和透射电镜观察β细胞结构的变化。结果HFSD组小型猪PG和Ins水平增高,以及加重胰岛素抵抗(IR)和β细胞损害。HFSD+1%Ibrolipi m组FPG显著降低,PG清除率以及胰岛素分泌急性相都明显改善,β细胞的结构和功能受到保护。结论Ibrolipi m有降低PG,减轻IR和保护β细胞的作用。Objective To explore the effect of ibrolipim on the structure and function of the islets in diabetic miniature pigs. Methods Fifteen Guizhou miniature pigs were randomly divided into 3 groups with similar body weight: normal control group (control diet, CD), experiment group (high fat and high sucrose diet, HFSD), treatment group (high fat and high sucrose diet plus treatment by 1% ibrolipim, HFSD + 1% ibrolipim). Blood samples were withdrawn from the orbital sinus of the animals at the end of each month after overnight fasting. The fasting concentrations of blood glucose and insulin were determined. Oral glucose tolerance test (OGTT) and insulin sensitivity test were performed at the end of month 6. The animals were sacrificed at the end of month 8. The pancreas tissues were dissected from each animal. The concentrations of insulin in pancreas tissues were analyzed by radio-immunity and BCA methods. The effects of ibrolipim on morphology of pancreas were observed with HE and immuno-histological stains and observed by transmission electron microscopy. Results Feeding a high-fat and high-sucrose diet to miniature pigs increased fasting plasma levels of glucose and insulin significantly. This diet also induced insulin resistance and impairment of pancreatic islet β cells. Supplementing of ibrolipim into high fat and high sucrose diet induced a decrease of concentration in plasma glucose, compared with swines fed with the sole high fat and high sucrose diet (P〈 0.05) and significantly improved clearance of plasma glucose and impairment of acute insulin response to glucose loading. The microstructure, ultrastructure, and immuno-staining results of β cells showed that the high fat and high sucrose feeding damaged the structure and function of the β cells. But ibrolipim had a protection of the structure and function of β cells. Conclusions Ibrolipim can alleviate insulin resistance, protect the structure and function of pancreatic islet β cells, and improve the glucose metabolism.
