摘  要:目的:观察红霉素对上皮细胞凋亡相关基因Bax和Bcl-2表达的影响,进一步探讨上皮细胞凋亡在鼻息肉组织形成中的意义。方法:将30例鼻息肉组织及6例下鼻甲组织的上皮细胞均分成两组,其中一组加红霉素(红霉素组),进行原代上皮细胞的培养,另一组加蒸馏水(对照组),以免疫组化法检测细胞凋亡基因Bax、抑凋亡基因Bcl-2。结果:鼻息肉上皮细胞Bcl-2和Bax表达均显著强于下鼻甲(均P<0.01);培养3d后鼻息肉上皮细胞红霉素组Bax表达显著强于对照组(P<0.05),Bcl-2表达虽强于对照组,但差异无显著性;下鼻甲上皮细胞红霉素组与对照组的Bcl-2与Bax阳性表达差异均无显著性(P>0.05);红霉素干预5d后鼻息肉上皮细胞显示明显的促凋亡趋势(P<0.05)。结论:红霉素能显著提高Bax蛋白的表达,有显著的促鼻息肉上皮细胞凋亡的作用。Objective To study the effect of erythromycin on the expressions of apoptosis-related genes Bax and Bcl-2 on the epithelial cell and further explore the significance of the cell apoptosis in the formation of nasal polyps. Methods The epithelial cells from 30 nasal polyps tissues and 6 inferior turbinate were evenly divided into two groups, Primary culture of the cells with erythromycin was conducted in one group, and the culture of the cells with distilled water was conducted in the other group. Expressions of Bcl-2 and Bax were detected by immunohistochemistry. Results Expressions of Bax and Bcl-2 on the epithelial cell were significantly higher in nasal polyps than in inferior turbinate (P 〈 0.01 ). Bax expression on the cells cultured with erythromycin was markedly higher than that of controls 3 days after the culture (P 〈 0.05). There was no significant difference in expressions of Bax and Bcl-2 on the epithelial cells from inferior turbinate between the erythromycin group and the control group (P 〉 0.05). An obvious increase in apoptosis of the epithelial cells from nasal polyps occurred 5 days after the intervention of erythromycin (P 〈 0.05). Conclusion Erythromycin can evidently elevate the expression of Bax and plays a critical role in inducing apoptosis of the epithelial cell in nasal polyps.
