摘　　要：目的研究血小板衍生生长因子(PDGF)及其受体(PDGFR)在肝细胞肝癌(HCC)组织中的表达及其与临床病理指标的关系,以探讨其在HCC发生、发展及转移中的作用。方法应用免疫组化方法结合计算机图像分析技术检测HCC组织、癌旁肝组织和正常肝组织中PDGF-A、PDGF-B及PDGFR-α、PDGFR-β的表达并分析其与HCC临床病理指标的关系。结果(1)HCC组织中PDGF-A、PDGF-B及PDGFR-α、PDGFR-β的阳性表达率分别为36.4%(16/44)、72.7%(32/44)、13.6%(6/44)、31.8%(14/44),明显高于癌旁肝组织和正常肝组织。HCC组织中PDGF-B的阳性表达率明显高于PDGF-A(P<0.01),PDGFR-β的阳性表达率显著高于PDGFR-α(P<0.05)。(2)HCC组织中PDGF及其PDGFR的表达强度均显著高于癌旁肝组织和正常肝组织(P<0.01)。不同分化程度、病理分级及有无门静脉癌栓的HCC组织中PDGF-A、PDGF-B及其PDGFR-α、PDGFR-β的表达强度差异均有高度显著性(P<0.01)。结论PDGF及其PDGFR的表达强度与HCC的分级及有无门静脉癌栓相关,提示在PDGF及其PDGFR在HCC的发生、发展和浸润转移过程中可能发挥重要作用。Objective To investigate the expression of platelet-derived growth factor （PDGF） and platelet-derived growth factor receptor （PDGFR） in primary hepatocellular carcinoma （HCC） and its relationship with clinical pathological parameters,exploring its role in the onset,development,invasion and metastasis of HCC. Methods The expressions of the two subtypes of PDGF,PDGF-A and PDGF-B,and their receptors,PDGFR-α and PDGFR-β in HCC ,para-carcinoma liver and normal liver were detected by immunohistochemistry and image analysis technique, and its relationship with the clinical pathological parameters was analyzed. Results （1） The positive rates of PDGF-A,PDGF-B,PDGFR-α and PDGFR-βin HCC were strikingly higher than those in para-carcinoma liver and in normal liver tissues,which were 36.4 % （16/44）, 72.7 % （32/ 44）, 13. 6% （6/44）, 31. 8% （14/44）, respectively. The positive rates of PDGF-B and PDGFR-βin HCC were significantly higher than those of PDGF-A and PDGFR-α （P〈0. 01,P〈0. 05,respectively）. （2） The degree of expression of PDGF and PDGFR in HCC tissues was significantly higher compared with that in para-carcinoma liver tissues （P〈0.01）. In HCC tissues of different differential degree,different pathological grades and with cancer embolus in portal vein or not, evident difference was found in the degree of expression of PDGF and PDGFR （P〈0. 01）. Conclusions The degree of expression of PDGF and PDGFR strongly correlats with the grades and invasion of HCC,indicating PDGF and PDGFR may play an important role in the onset,development ,invasion and metastasis of HCC.
