华夏小葱制剂对脂肪肝大鼠抗脂质过氧化损伤及细胞因子损伤的影响

摘　　要：目的研究华夏小葱制剂对脂肪肝大鼠的防治作用及其机制。方法将60只SD大鼠随机分为空白对照组,模型对照组,华夏小葱制剂低、中、高剂量组以及东宝肝泰片组。用高脂饮食、酒精水并结合皮下注射小剂量四氯化碳的方法建立脂肪肝模型。结果模型对照组总胆固醇(TC)、甘油三酯(TG)、丙二醛(MDA)、核因子-κB(NF-κB)、转化生长因子-β(TGF-β)、单核细胞趋化蛋白-1(MCP-1) mRNA值显著高于空白对照组(P<0.05或P<0.01),超氧化物歧化酶(SOD)活力显著低于空白对照组(P<0.01);华夏小葱制剂低、中、高剂量组及东宝肝泰片组TC、TG、MDA、NF-κB、TGF-β、MCP-1 mRNA值显著低于模型对照组(P<0.05或P<0.01),SOD活力显著高于模型对照组(P<0.01)。结论华夏小葱制剂对脂肪肝大鼠具有防治作用,其机制可能与降低大鼠体内血脂、减少脂质过氧化损伤及细胞因子的损伤有关。Objective To study the preventive and therapeutic effect of Prepration of Chinese Allium fistulosum L. （PCAL） on fatty liver （FL） rats and its mechanism. Methods Totally 60 SD rats were randomly divided into blank control group, model control group, small, middle and large dosage of PCAL groups and Dongbao Gantai Tablet group. The rats were fed with high fat diet and water comprised of ethanol and were hypodermically injected with small dosage of carbon tetrachloride to establish FL model. Results As compared with blank control group, the content of total cholesterol （TC）, triglyceride （TG）, malondialdehyde （MDA）, nuclear transcription factor-κB （NF-κB）, transforming growth factor-β （TGF-β） and monocyte chemoattractant protein-1 （MCP-1） mRNA in model control group increased significantly （P〈0.05 or P〈0.01 ）, while the activity of superoxide dismutase （SOD） decreased significantly （PM0. 01）. As compared with model control group, the content of TC, TG, MDA, NF-κB, TGF-β and MCP-1 mRNA in small, middle and large dosage of PCAL groups and Dongbao Gantai Tablet group decreased significantly （P〈 0.05 or P〈 0.01）, while the activity of SOD increased significantly （P〈0. 01）. Conclusion PCAL has a protective and therapeutic effect on FL rats, which might be relevant to its reducing blood fat, diminishing damage of lipid peroxidation and cytokine.
