摘  要:目的观察大鼠暴露尼古丁对中脑腹侧背盖区(VTA)、黑质(SN)和大脑皮质α-7型神经烟碱胆碱能受体(α-7-nAChRs)表达的影响。方法建立暴露尼古丁的大鼠动物模型,应用免疫组织化学法、原位杂交法分别观察VTA、SN和大脑皮质α-7-nAChRs蛋白和mRNA表达的变化,用Western blotting方法分析尼古丁作用后PC12细胞α-7-nAChRs表达的变化。结果尼古丁上调大脑皮质α-7-nAChRs蛋白表达,也上调VTA和SNα-7-nAChRsmRNA的表达。在PC12细胞中,α-7-nAChRs表达随着尼古丁作用时间和剂量的增加而增加。结论尼古丁在转录水平上调VTA和SNα-7-nAChRs的表达,在翻译水平上调大脑皮质α-7-nAChRs的表达。Objective To observe the regulation of α-7 neuronal nicotinic acetylcholine receptors (α-7-nAChRs) by nicotine in the ventral tegmental area(VTA), the substantia nigra (SN) and the cortex of the rat. Methods Nicotine exposed animal models were established. The changes of α-7-nAChRs proteins and mRNA were observed by immunohistochemistry and in situ hybridization in VTA, SN and cortex, and the expression of α-7-nAChRs proteins by Western blotting in PC12 cells. Results The expression of α-7-nAChRs proteins in cortex and mRNA in VTA and SN were upregulated by nicotine, The expressions of α-7-nAChRs in PC12 cells were in proportion to and dependent on the time for nicotine to function and the dose. Conclusion Both α-7-nAChRs mRNA in VTA and SN in transcriptional mechanism and α-7-nAChRs protein in the cortex at the translation level were upregulated.
