摘  要:以铂电极为基底,用恒电位沉积一层普鲁士蓝(PB),然后用纳米金和明胶构成的复合固酶基质,结合溶胶凝胶技术将辣根过氧化物酶(HRP)固定在普鲁士蓝修饰的电极表面,再用戊二醛(GA)对复合酶膜进行交联改性,从而制备了性能良好的过氧化氢传感器.采用循环伏安法(CV)、计时电流法、交流阻抗法(EIS)对传感器进行了电化学表征.在pH=6·0的磷酸盐缓冲溶液(PBS)中,探讨了工作电位、pH、温度以及干扰物质对传感器响应电流的影响.实验结果表明,此方法有较高灵敏度、较好抗干扰能力、良好稳定性及重现性.传感器对H2O2的线性响应范围为1·2×10-6~1·3×10-3mol/L,检测限为6·0×10-7mol/L.A novel type of hydrogen peroxide biosensor was prepared. A stable Prussian Blue (PB) film was electrodeposited on platinum electrode by applying constant potential. After that, horseradish peroxidase (HRP) was entrapped to colloidal gold combined with the gelatin, which were immobilized onto the surface of Prussian Blue modified platinum electrode. Then the HRP-nano-Au-gelatin film was treated with glutaraldehyde (GA). Cyclic vohammetry (CV), electrochemical impedance spectroscopy (EIS) and chronoamperometry (CA) were used to characterize the electrochemical properties of the biosensor. The factors influencing the performance of the biosensor (applied potential, pH, temperature and interfering substances) were investigated in 0.05 mmol/L phosphate buffer (pH=6.0). The biosensor exhibited fine selectivity, good repeatability, high sensitivity and good stability. The linear response of the biosensor to hydrogen peroxide is in the concentration range of 1.2 ×10^-6~1.3×10^-3 mol/L with a detection limit of 6.0×10^-7 mol/L.
