摘 要:目的初步探索职业性慢性铅中毒患者T淋巴细胞亚群及Th淋巴细胞因子的变化特点。方法对23例职业性慢性铅中毒患者(铅中毒组)及20例健康非职业铅接触成人(对照组)采用流式细胞技术检测外周静脉血淋巴细胞CD_3、CD_4、CD_8的表达,同时通过流式细胞微球芯片捕获技术检测血浆IL-2、IL-4、IL-6、IL-10、TNF-αt、IFN-γ的浓度。结果铅中毒患者外周静脉血CD_4相对百分比(32.68%±11.54%)及CD_4/CD_8比值(0.89±0.39)与对照组比较,明显下降,差异有统计学意义(P<0.05);血浆IL-2浓度为(2.00±0.68)pg/ml,与对照组比较明显上升,差异有统计学意义(P<0.05);血浆IL-10和IFN-γ浓度分别为(1.83±0.85)pg/ml和(3.42±0.85)pg/ml,与对照组比较明显减低,差异有统计学意义(P<0.05)。结论T淋巴细胞亚群及细胞因子对评价铅免疫毒性有进一步研究价值。Objective To investigate the changes in T lymphocyte subsets and plasma Th1/Th2 cytokine levels in patients with occupational chronic lead poisoning. Methods Flow cytometric analysis was used to count the numbers of CD3^+, CD4^+, CD8^+ cells and cytometric bead array (CBA) capture technique was used to quantify plasma IL-2, IL-4, IL-6, IL-10, TNF-α, IFN-γ concentrations. Twenty-three patients with occupational chronic lead poisoning and 20 controls were examined. Results Compared with control group, CD3^+ CD4^+ lymphocytes and CD4/CD8 ratios decreased in patients with occupational chronic lead poisoning. The plasma IL-2 level were significantly raised while the IL-10 and IFN-γ were reduced in patients with occupational chronic lead poisoning. Conclusions T lymphocyte subsets and plasma Th1/Th2 cytokine levels could be a new parameter to assess lead immunotoxity.
