摘 要:目的:建立测定鬼臼毒素微乳凝胶剂中鬼臼毒素含量的方法。方法:采用反相高效液相色谱法,色谱柱为Lichrospher C_(18)(250 mm×4.6 mm,5μm),以甲醇-水(63:37)为流动相,流速为0.8 mL·min^(-1),检测波长为292 nm。结果:鬼臼毒素在200~800μg·mL^(-1)范围内呈良好的线性关系,r=0.999 9。平均回收率为99.6%,RSD为0.6%;精密度与溶液稳定性RSD分别为0.2%和0.2%。结论:方法快速,重现性、精密度及准确度均能满足鬼臼毒素含量测定要求。Objective: To establish a RP-HPLC method for the determination of podophyllotoxin in microemulsion-based gels. Methods: The RP-HPLC assay was conducted on a Lichrospher C18 column (250 mm × 4.6 mm, 5μm) with a mobile phase composed of methanol:water (63:37 ) with a flow rate of 0.8 mL· min ^- 1 and detection at 292 nm. Results : The linearity of podophyllotoxin concentration curve was in arange of 200~800μg·mL^-1(r =0.9999). The average recovery was 99.6% with a RSD of 0.6%. The RSDs of the accuracy and solution stability were 0.2% and 0.2% , respectively. Conclusion : This rapid, accurate and reproducible RP-HPLC method was useable for quantification of podophyllotoxin in microemulsion-based gels.
