感染亚洲带绦虫的家猪血清IL-2和INF-γ含量变化的意义

摘　　要：目的研究家猪感染亚洲牛带绦虫后血清细胞因子IL-2和INF-γ的含量变化,从细胞因子角度探讨家猪抗亚洲牛带绦虫免疫应答的机制。方法用亚洲牛带绦虫虫卵直接灌胃20d龄健康乳猪5头,并以健康乳猪4头作对照。于感染后第7、13、15、30、41、45、60和75天,定期抽取猪心脏血并分离血清。用ELISA试剂盒检测血清细胞因子IL-2和INF-γ含量。结果正常组血清IL-2和INF-γ含量在不同时期基本处于稳定水平;实验组血清IL-2和INF-γ含量在感染第7天时含量最高,之后迅速下降略高于或低于正常组,第60天开始回升,第75天形成新的高峰。与正常组相比,实验组血清IL-2和INF-γ含量在第7、75天显著升高(P<0.01),在感染第13、60天时略升高(P<0.05)。结论家猪感染亚洲牛带绦虫不同时期,血清IL-2和INF-γ含量存在较大差异。感染早期血清出现高水平IL-2和INF-γ,提示细胞免疫在抗击六钩蚴早期感染中发挥着重要作用;感染中期,血清IL-2和INF-γ含量相当于正常组,提示细胞免疫应答水平低下;晚期出现高水平的IL-2和INF-γ,可能是宿主抑制囊尾蚴引起肝纤维化形成的重要因素。Objective To detect the levels of serum IL-2 and INF-γ in domestic pigs experimentally infected with Taenia saginata. Methods Five helthy pigs were fed with eggs of Taenia saginata, and reared isolately in animal center to establish Taenia saginata infection model, and normal controls. Blood samples were obtained at day 7, 13, 15, 30, 41, 45, 60 and 75 after infection. Serum IL-2 and INF-γ were detected by ELISA. Results The levels of serum IL-2 and INF-γof the normal group maintained relatively stable through out the experiment. The levels of serum IL-2 and INF-γof the infected group were peaked at day 7, and then decreased gradually, but went up again at day 60 and peaked at day 75. Compared with the normal group, the levels of serum IL-2 and INF-γof the infected group showed a great increase at day 7 and day 75, showed a moderate increase at day 13 and day 60. Conclusion The levels of serum IL-2 and INF-γshowed a biphasic change at difference infection stages. Hight levels of serum IL-2 and INF-γat early infection stage suggested that cellular immune response played very important role against Taenia saginata at early infection stage. IL-2 and INF-γmight be important factors in inhibiting liver fibrosis in the late infection stage.
