自然流产患者蜕膜MMP10的表达及临床意义

摘　　要：目的:探讨基质金属蛋白酶10(MMP10)基因及其蛋白在人自然流产蜕膜中的表达水平及临床意义。方法:利用RT-PCR和Western blotting技术分别检测MMP10基因和蛋白在人正常蜕膜和自然流产蜕膜的表达水平、利用免疫组织化学技术检测MMP10在人正常蜕膜和自然流产蜕膜组织中的分布。结果:正常蜕膜中MMP10基因(0.20±0.08)及蛋白(0.25±0.07)表达均较低,在自然流产蜕膜中MMP10基因(0.65±0.10)及蛋白(0.79±0.10)表达均较高,两者差异具有显著性意义(P<0.01);MMP10主要表达在流产蜕膜的基质细胞。结论:MMP10在流产蜕膜中高表达可能有利于降解细胞外基质,促进胚胎滋养细胞的侵袭,从而延缓自然流产。Objective：To discuss expression levels of MMP10 gene and protein in human spontaneous abortion deciduas and its significance in clinic. Methods：Expression levels of MMP10 gene and protein in human spontaneous abortion decidua were determined by RT-PCR, Western blotting and distribution by immunohistochemistry. Results：Expression levels of MMP10 mRNA and protein in all normal deciduas were lower （0.20 ±0.08,0.25 ±0.07）,but all increased in Spontaneous Abortion Deciduas （0.65 ±0.10,0.79 ±0.10 ）. The difference between the two groups was significant （ P〈0.01, MMP 10 was mainly expressed in stromal cells.Conclusion：The results suggested that higher expression of MMP10 in spontaneous abortion deciduas might resolve the matrix metalloproteinase,promote the extrovillous cytocyst cells invasion and prohibit the spontaneous abortion.
