摘  要:目的:研究CD40-CD40L系统在兔腹主动脉粥样硬化斑块进展过程中的表达以及与斑块稳定性的相关性。方法:50只健康新西兰大白兔,随机均分为10组:正常对照1~5组(C1~5组)、动脉粥样硬化模型1~5组(AS1~5组)。C1~5组分别予兔标准饲料喂养至8周、10周、12周、14周、16周取腹主动脉标本;AS1~5组均在高脂饲养1周后予腹主动脉球囊导管损伤术造模,并分别持续高脂饮食至8周、10周、12周、14周、16周取标本。各组均于实验前及取标本前检测血清C反应蛋白(CRP),各组腹主动脉标本通过免疫及特殊染色定位、光镜下摄片并计算机图像分析软件测定斑块内CD40、CD40L、MMP-1阳性染色的面积占血管内膜面积百分比值及胶原/脂质含量比值,分析CD40-CD40L系统的表达与斑块稳定性的相关性。结果:AS1~5组均形成腹主动脉粥样硬化斑块。与C1~5组对应比较,AS1~5各组血清CRP均显著增高(P<0.01),斑块内CD40、CD40L、MMP-1的表达显著增加(P<0.01)。在动脉粥样硬化进程中,CD40、CD40L表达与CRP、MMP-1表达呈正相关,与胶原/脂质含量比值负相关。结论:斑块中CD40-CD40L系统表达随着动脉粥样硬化进展而逐渐增高,其高表达与斑块的不稳定性增加密切相关。Objective:To study the expression of CD40-CD40L system on rabbit aorta plaque and correlation with plaque stability during atherosclerotic process. Methods:50 healthy rabbits were devided into 10 groups in random:five controlled groups (C1-5 group)and five AS model groups (AS1-5 group). Rabbits in C1-5 group were fed on regular diets until 8 weeks,10 weeks.12weeks,ldweeks and 16weeks to get the aorta specimen respectly.Correspondingly rabbits in AS1-5 groups were given high lipid diets continously until 8weeks, 10weeks, 12weeks, 14weeks and 16weeks,during which each AS group was conducted balloon injury at the end of the first week to promote the establishment of AS models. All the groups were detected serum CRP individually at beginning and before specimen obtained.Immunohistochemistry and special stain was used to detect the intima expression proportion of CD40,CD40L,MMP-1 as well as contant of collagen and lipid photographed under microscope and measured by computer.Then the collagen/lipid ratio were calculated.Finally correlation between CD40-CD40L expression and plaque stability were analyzed. Results:All atherosclerotic plaques were successfully established on all rabbits'abdominal aorta in the 5 AS groups.Compared with C1-5 groups correspondantly, the expression of CD40,CD40L,MMP-1 were significantly increased (P〈0.01)in every AS model group.Meanwhile serum CRP levels were higher than those of each C group(P〈0.01)evidently. The correlation between CD40-CD40L system and CRP as well as MMP-1 is positive.While negative correlation is between CD40-CD40L level and collagen/lipid ratio. Conclusion:The expression levels of CD40-CD40L on plaque lesions were increasingly high with atherosclerusis processing.High expression of CD40-CD40L system has close relation to plaque vulnerability.
