摘 要:目的建立蒙山九州虫草菌丝体和发酵液多糖水解液组分的定性分析方法。方法蒙山九州虫草菌丝体和发酵液多糖水解成单糖后,用α-萘胺进行衍生,得到α-萘胺衍生单糖。采用50μm(i.d.)×57 cm未涂层石英毛细管,以75 mmol/L硼砂(pH 10.5)为背景电解质,于200 nm处进行紫外检测,操作温度25℃,分离电压8 kV。采用标准品对照进行定性分析。结果蒙山九州虫草发酵液多糖和菌丝体多糖水解后主要含有葡萄糖、甘露糖和半乳糖,另外还含有少量阿拉伯糖。结论该方法样品用量少、时间短、操作简单,可以作为单糖定性分析的常规方法。Purpose To develop a capillary zone electrophoresis method for qualitative determination of polysaccharides from the mycelium and zymolytic liquid of Cordyceps kyushuensis. Methods Polysaccharides from the mycelium and zymolytic liquid of Cordyceps kyushuensis were hydrolyzed into monosaccharides, which were further derived with α-naphthylamine. The CZE determination was performed on an uncoated fsedsilica capillary column with 50 μm (id), using 75 mmol/ L sodium tetraborate buffer (pH 10.5), UV detector at 200 nm, operation voltage 8 kV and temperature 25 ℃. Standard monosaccharide were used for reference. Results The monosaccharides composition of polysaccharides was obtained. The results showed that the main components were glucose, galactose and mannose. Conclusions The developed CZE method is simple, convenient and useful for qualitative determination of monosaccharides.
