银杏叶对成人视网膜神经细胞活性的影响

摘　　要：目的研究银杏叶对视网膜神经细胞活性的影响,为治疗退行性视网膜疾病等提供参考依据。方法以成人视网膜为研究对象,以24h和72hMTT检测,透射电镜超微形态学观察,细胞线粒体膜电位测定等为研究手段,研究银杏叶对成人视网膜神经细胞活性的影响。结果细胞超微形态学观察发现,银杏叶药物组培养2周的细胞外节段盘膜排列规整,内节段线粒体丰富,细胞核染色质均匀。MTT检测发现,银杏叶能够较好地促进视网膜神经细胞增生,且以15.625mg/ml时保护作用最明显。银杏叶能够显著升高视网膜神经细胞的膜电位并使兴奋性呈波动性变化。结论银杏叶对培养的视网膜神经细胞有良好的保护作用。Objective Ginkgo biloba was generally used in clinic but seldom in therapy of retinal diseases. Human retinal nerve cells were difficult to be obtained. This study aimed to show the possibly protective effects of Ginkgo biloba on cultured human retinal nerve cells. Methods Six corpse retinas aged 32. 2 ± 1.4 years from Tongren eye bank were obtained. Retinal nerve cells were co-cultured with Ginkgo biloba. 24 hours and 72 hours later, MTT assays were used to evaluate the proliferation of retinal nerve cells. The uhrastrnctural alternation of cultured cells was observed with transmission electron microscopy （TEM）. Confocal microscopy was utilized to evaluate mitochondrial membrane potentials （MMP） of cultured cells. Results Co-cultured retinal nerve cells with Ginkgo biloba were shown survival well under the TEM. The discs of outer segments of photoreceptor remained a good morphology and displayed a regulate arrangement, and plentiful chondrosomes of inner segments and well- distributed nuclear chromatins were seen after two weeks. The addition of Ginkgo biloba promoted survival of adult retinal neurons significantly in concentration-dependent manner in 1. 953 - 15.625 mg/ml with the strongest effect in 15. 625 mg/ml. The MMPs were significantly enhanced after addition of Ginkgo biloba in comparison with before administration of Ginkgo biloba. Conclusion Ginkgo biloba can protect cultured retinal nerve cells in vitro.
