乌司他丁对大鼠移植小肠缺血再灌注损伤的保护作用

摘  要:目的评价乌司他丁对大鼠移植小肠缺血再灌注损伤的保护作用。方法将受体分为三组:(1)对照组A:两人同时操作,将刚离体的供肠立即植入受体,使移植肠的冷缺血时间为0;(2)对照组B;(3)UTIB组。后两者按照正常的单人操作,供肠的冷缺血时间平均为1h,第1组的术后处理同第2组,即不使用任何药物干预。分别于刚离体时、术后4、24h切取供肠3cm,测定其中丙二酰二醛(malondialdehyde,MDA)的含量。结果与各自的基础含量相比,三组供肠组织中的MDA含量在供肠血运恢复后的第4、24h均明显地升高并维持在一个较高的水平(P<0.05)。与其他两组相比较,对照组B升高最显著(P<0.05),而UTIB组则在上述2个时点的MDA含量与对照组相似(P>0.05)。结论酶抑制剂UTI能抵抗缺血再灌注时氧自由基(oxygen free radiacls,OFRs)所致的损伤而具有保护作用,从而促进移植肠结构和功能的恢复。Objective To evaluate the protective effects of Ulinastatin (UTI) against ischernia- reperfusion injury of the graft in rat small bowel transplantation(SBT). Methods We divided recipients into 3 goups in random: 1. control goup (0), the goup of zero- cold - ischemia models were made that the grafts were transplanted as soon as they were removed from the donor rats under the collaboration of two operators. 2. control group ( 1 ), 3. UTI group ( 1 ). These former two groups were not interfered with any drug after SBT. Then regarded the control group (1) and the UTI group ( 1 ) of the former experiment as another 2 groups. Because the cold - ischemia - time of the grafts in these 2 groups were meanly lh under one operator manipulating in ordinary. 3era length of distal small bowel was acquired from each graft as soon as it was removed from the donor, measuring MDA content as base content (control). Then same length of graft was harvested from each recipient at 4 and 24h after revasculaxization respectively. All MDA contents were measured by commercial kits. Results All MDA contents of 3 groups obviously increased and maintained a high level than their base contents ( P 〈0. 05) during the early post - revascularization period (within first 24h). Among three groups, control( 1 )group showed the highest elevation, and its MDA content was greatly higher than that of the other groups ( P 〈 0,05). and the MDA content of UTI group ( 1 ) was the same as that of control group (0) at 4h and 24h respectively after revascularization ( P 〉 0.05). Conclusions It is supposed that UTI demonstrates protective effects against ischemia - repefusion injury of OFRs ( oxygen free radicals) in rat SBT, and helps to the structural and functional recovery of the transplanted small bowl.
