摘 要:目的:改良去细胞异体神经的制备方法,研制出一种理想的自体神经移植替代物.方法:将用Sondell法(Triton X-100、脱氧胆酸钠)和改良法[Triton X-200,sulfobetaine-10(SB-10),sulfobetaine-16(SB-16)]两种方法制备的去细胞异体神经用HE染色、髓鞘染色、免疫组化染色、扫描电镜检查等进行组织学评价.然后将制备的神经桥接大鼠坐骨神经缺损,从坐骨神经指数、神经电生理、运动终板染色等方面评价神经移植后的再生效果.结果:改良法制备的神经细胞及髓鞘去除彻底、结构保存完好;移植后神经再生良好,优于Son-dell法制备的神经,达到了与自体神经移植相当的再生效果.结论:综合运用萃取剂Triton X-200,SB-10和SB-16的改良化学萃取方法是一种较为理想的去细胞异体神经制备方法,其制备的异体神经移植物可望替代自体神经移植.AIM: To improve the preparative method of acellular nerve allograft and to develop an ideal substitute for autogenous nerve graft. METHODS- Rat sciatic nerves were chemically extracted by Sondell method, with detergents of Triton X-100 and sodium deoxycholate, and by improved method, with detergents of Triton X-200, sulfobetaine-10 (SB-10) and sulfobetaine-16 ( SB- 16). The acellular nerve allografts prepared were evaluated by HE staining, myelin staining, immunohistochemistry and scanning electron microscopy. Rat sciatic nerve defects were bridged by the allografts and autografts. The outcomes of nerve regeneration were evaluated by sciatic nerve function index ( SFI), nerve electrophysiology, and motor end-plate staining. RESULTS: The cells and myelin were removed thoroughly and the nerve structures were preserved well in acellular nerve allografts prepared by improved method. After grafting, more satisfactory effects of nerve regeneration were achieved in improved method group compared with Sondell method group. And the effects of nerve regeneration in improved group were similar to those in autograft group. CONCLUSION: This new chemical extraction procedure with detergents of Triton X-200, SB-10 and SB-16 is an ideal paradigm to develop high-quality acellular nerve allograft. And this acellular nerve allograft is promising to replace the autologous nerve graft.
