加味桃核承气汤含药血清对胰岛素诱导兔血管平滑肌细胞增殖的抑制作用

摘　　要：目的:探讨加味桃核承气汤含药血清对胰岛素诱导兔血管平滑肌细胞增殖的抑制作用。方法:采用兔主动脉平滑肌细胞株(SMC),以胰岛素(RI)诱导SMC增殖,制备桃核承气汤不同配伍组方含药血清,MTT法观察各组对RI诱导SMC增殖的抑制作用,以罗格列酮及美迪康作为对照组,探讨各组在糖尿病大血管病变发生与发展中可能的作用。结果:RI诱导SMC增殖试验确定RI终浓度为300mu/L;加味桃核承气汤含药血清对RI诱导SMC增殖的抑制作用试验显示,活血+泻热通下组、桃核承气汤组、罗格列酮组增殖抑制作用十分显著(P<0.01),以活血+泻热通下组抑制率最高(39.8%)。美迪康组、泻热通下组及益气养阴组增殖抑制作用不明显(P>0.05)。结论:加味桃核承气汤可能通过抑制RI诱导SMC增殖作用抗糖尿病动脉粥样硬化所致的大血管病变,本方抑制平滑肌细胞增殖的有效成份可能存在于活血+泻热通下组中。To discuss the inhibitory action of insulin-induced smooth muscle cell proliferation by medicated blood serum of Jiawei Taohe Chengqi Decoctions. Methods ： Insulin was applied to aortic smooth muscle cell of rabbit to induce cell proliferation and prepare medicated blood serum of Jiawei Taohe Chengqi Decoctions. Rosiglitazone and mefformin hydrochlofide were used respectively to establish control groups. We observed the inhibitory action of insulln-induced smooth muscle cell proliferation by medicated blood serum with the method of methythiazolyl tetrazolium assay. Results： The final concentration of insulin was 300 mu/L. Significant inhibitory actions of smooth muscle cell proliferation were observed in three groups, which were group of Taohe Chengqi Decoction, group of promoting blood circulation plus purging heat, and group of rosiglitazone （P 〈 0.01）. Among the three groups, the group of promoting blood circulation plus purging heat showed the highest inhibition rate of 39.8 %. However, there was no significant inhibitory action of smooth muscle cell proliferation in the group of mefformin hydrochloride, the group of purglng heat, and the group of Yiqi Yangyin （ P 〈 0.05）. Conclusion： Jiawei Taohe Chengqi Decoction has an effect of anti-macrovascular lesion in type Ⅱ diabetes. The active ingredient of anti smooth muscle ceil proliferation might lie in the group of promoting blood circulation plus puring heat.
