摘  要:目的 探讨冻融和移植过程对人胎儿卵巢组织超微结构的影响。方法 6例23~34w引产的胎儿卵巢慢冻速融,并移植到裸鼠肾被膜下,分别于移植后48h、7和28d回收移植物。卵巢组织分为新鲜对照组、冻融组、移植后48h、7d及28d组。电镜和光镜下观察各组形态学改变。结果胎儿卵巢组织冻融前后光镜下的卵泡密度(23.92±3.94)及(22.70±4.18)无显著差异,移植后48h卵泡密度(16.70±3.98)与移植前比较显著下降(P〈0.01)。各组卵泡超微结构整体上保存良好。冷冻对卵巢间质损伤较大且局部差异大,移植后48h间质细胞稀疏,白细胞浸润;移植后7及28d,间质内可见较多胶原纤维束,大量微小血管生长。结论人胎儿卵巢组织的冻融损伤主要表现在间质,而移植损伤在移植后48h内最严重;结合光镜和电镜观察手段能更全面评价冻融过程和移植过程对胎儿卵巢组织的影响。Objective. To investigate the effect of cryopreservation and transplantation on the histological appearance and uhrastructural morphology characteristics of human fetal ovarian tissue. Methods: Ovarian tissues from induced aborted fetus aged 23 w-34 w (n= 6) were cryopreserved, rapidly thawed and xenografted into the nude mice, then the grafts were recovered at 48 h, 7 d and 28 d respectively. Ovarian cortex samples were processed for light and transmission electron microscopy (TEM). Results: Histological analysis revealed that no significant differences in follicle count was found between the fresh group and the frozen-thawed group (23. 92±3.94 vs 22.70±4. 18). Follicle counts at 48 h after transplantation were significantly decreased(16.70±3.98)compared with those of the fresh group and the frozen-thawed group (P〈0. 01). Follicle ultrastructure was generally well preserved in all groups, while the stroma showed variable preservation. Matrix density decreased 48 h after transplantation, and the stroma was filled with collagen bundles and numerous microvessels were found in the grafts in 7 d and 28 d groups. Conclusions: The stroma of human fetal ovarian tissues was found to be sensitive to cryoinjure, and the transplantation damage was showed to happen mainly within the first 48 h after grafted. Light microscopy and TEM could be a useful tool for evaluation of the effect of cryopreservation and transplantation.
