摘 要:目的:观察大鼠正畸牙齿移动过程中基质金属蛋白酶-2(MMP-2)的表达和分布变化。方法:选用40只雄性SD大鼠,于加力后0 d、3 d、7 d、14 d、21 d处死大鼠,免疫组化法检测明胶酶-2(MMP-2)的表达,计算平均光密度(OD)和积分光密度(IOD)。结果:牙齿移动后牙周膜两侧MMP-2表达增加,破骨细胞、破牙本质细胞、骨细胞、成纤维细胞和部分成骨细胞、成牙本质细胞染色阳性。OD表现动态变化,7 d最低,而后缓慢升高。加力后IOD逐渐升高,7 d达到顶峰而后缓慢下降,21 d时仍维持在高水平。结论:牙齿移动时MMP-2呈规律性变化,与牙齿正畸骨改建关系密切。Objective: To observe the expression and distribution of gelatinase-A (MMP-2) in the periodontal tissues of rats during tooth movement. Methods: Forty male Sprague-Dawley rats were used. The animals were sacrificed after 0d, 3d, 7d, 14d and 21 days respectively. Immunohistochemistry was applied to localize and examine the expression of gelatinase-A (MMP-2) in periodontal tissue. The average optical density (OD) and integrated optical density (IOD) were calculated. Results: In tooth movement groups, the MMP-2 expression increased and was observed on both sides periodontium of movement tooth. Osteoclast, cementoblast, osteocyte, fibroblast, a part of osteoblast and cementoblast appear positive. Dynamic changes of OD occurred. OD reached the minimum on the 7th day and then increased slowly. IOD increased steadily, up to the peak on the 7th day, and then decreased, which still remained a high level on the 21 st day. Conclusion:MMP-2's activity changes regularly, in close relation with bone remodeling during the orthodontic tooth movement.
