摘  要:[目的]同时测定亚急性衰老小鼠脑皮层一氧化氮(NO)含量和一氧化氮合酶(NOS)活力,研究NO和NOS与衰老的关系。[方法]造模组连续42天于小鼠颈背部注射D-半乳糖复制亚急性衰老小鼠模型;对照组以生理盐水注射。用硝酸还原酶法测定脑皮层NO含量;采用化学比色法测定脑皮层NOS活力。[结果]D-半乳糖诱导的亚急性衰老模型组大小脑的NO含量较对照组显著增加;D-半乳糖诱导的亚急性衰老小鼠组大脑的NOS活力较正常组也有升高趋势。[结论]D-半乳糖致亚急性衰老小鼠脑皮层NO含量和NOS活力增加与衰老发生关系密切,可以将NO含量和NOS活力变化作为观察神经系统衰老的敏感指标。[Objective] To investigate relationships between NO, NOS and senescence by simultaneous determination of NO content and NOS activity. [Methods]Nuchal injection of D-galactose was administered continuously for 42 days to produce subcute senescence models, whereas normal saline for the control group. The NO content in in cerebral cortex was determined by nitrate reduetase method and the NOS activity was detected by chemical colorimetry. [Results]Contents of NO in cerebrum and cerebellum in the model group significantly increased compared with the control group; NOS activity in cerebrum in the model group was more intense than that in the control group. [Conclusion]D-galactose-induced increases of NO content and NOS activity are closely correlated with the development of senescence. Changes of NO content and NOS activity can be a sensitive index of neural aging.
