摘  要:目的 探讨染料木素(genistein)体外抑制人胃癌BGC~823细胞生长及诱导其凋亡的作用机制。方法 采用四甲基偶氮唑盐(MTT)法检测染料木素对人胃癌BGC-823细胞的增殖抑制效应;DAPI荧光染色观察细胞凋亡形态;流式细胞仪检测染料木素作用24h后BGC-823细胞的周期分布和凋亡率;RT—PCR方法检测环氧合酶-2(COX-2)在mRNA水平的表达;进一步采用Westernblotting法检测COX-2在蛋白质水平的表达。结果 染料木素对BGC-823细胞具有增殖抑制效应,并有时间及浓度依赖性。染料木素作用24、48、72h的半数抑制浓度(IC50)分别为(81.04±1.03)、(31.85±2.26)、(20.73±2.11)μg/mL。10~80μg/mL染料木素作用24h后,BGC-823细胞出现核碎裂,呈现典型的凋亡特征,细胞被阻滞于G2/M期,随药物质量浓度的增加,细胞凋亡率逐渐增加,细胞内COX-2表达在mRNA水平和蛋白质水平均下调。结论 染料木素对人BGC-823细胞有较强的抑制增殖和诱导凋亡作用,其机制可能与抑制COX-2表达有关。Objective IO investigate the effect of genistein on COX-2 expression in human gastric cancer BGC-823 cells and to explore its potential mechanisms of inhibiting proliferation and inducing apoptosis. Methods MTT Assay was used to observe the effect of genistein after 24 h on proliferation of BGC-823 cells. Cell cycle and apoptosis were analyzed by flow cytometry (FCM) and DAPI staining. Transcription level of COX-2 was detected by RT-PCR and protein level of COX-2 was detected by Western blotting. Results Genistein significantly inhibited the proliferation of BGC-823 cells in dose- and time-dependent manners, the IC50 value of genistein at 24, 48, and 72 h were (81.04±1.03), (31.85± 2.26), and (20. 73±2.11) vg/mL, respectively. When treated with 10-80 μg/mL genistein for 24 h, BGC-823 cells were arrested at G2/M phase, and the apoptotic rate was increased along with the increase of genistein concentration, meanwhile, COX-2 expression at transcription and protein level was decreased. Conclusion Genistein has a stronger inhibition on proliferation and induction on apoptosis of BGC-823 cells through inhibiting COX-2 expression.
