消脂护肝胶囊对非酒精性脂肪肝大鼠病理及CYP2E1基因的影响

摘　　要：目的:研究消脂护肝胶囊对非酒精性脂肪性肝病(NAFLD)大鼠模型肝脏病理和肝细胞色素P4502E1(CYP2E1)基因表达的影响。方法:32只SD大鼠随机分为空白组、模型组、观察组和对照组4组,后3组以高脂饲料联合四环素腹腔注射建立NAFLD模型,造模1周后分别给予消脂护肝胶囊和东宝肝泰片治疗。实验结束时观察各组肝匀浆游离脂肪酸(FFA)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量、肝脏病理及肝组织CYP2E1 mRNA表达情况(RT-PCR法)的变化。结果:观察组肝匀浆FFA、MDA含量降低,SOD增加,肝脂肪变性程度减轻,CYP2E1 mRNA表达减少,效果优于东宝肝泰对照组。结论:消脂护肝胶囊能够显著抑制CYP2E1 mRNA表达,阻止脂质过氧化反应,从而发挥防治NAFLD的作用。Objective： To study the effect of XiaozhiHugan capsule on pathologic changes and cytochrome P450 2El （CYP2E1） gene expression of rats with non-alcoholic fatty liver. Methods：32 male SD rats were divided into four groups randomly： rats in the normal group, the model group, the observe group and the DongBaoGanTai controlled group. The last three groups were given high-fat diet associated with peritoneal injection of tetracycline to set up NAFLD models. After exposure to high-fat diet associated with peritoneal injection of tetracycline for 1 week, the observe group and control group were treated by corresponding medicines. Changes of Hepatic CYP2E1 mRNA expression （RT-PCR）, patholog, contents of free fatty acid （FFA）, malondialdehyde （MDA） and superoxide dismutase （SOD） in liver homogenate in different groups were detected in the end. Results： In the observe group hepatic CYP2E1 mRNA expression, the contents of FFA, MDA in liver homogenate was decreased and SOD increased. The degree of fat degeneration was meliorated, which has so much superiority more than that of in Dong- BaoGanTai control group. Conclusions： XiaoZhiHuGan capsule plays an important role oin preventing and treating NAFLD by inhibiting the expression of CYP2E1 mRNA and the reaction of lipid peroxidation remarkably.
