摘 要:目的研究Jun活化结构域结合蛋白-1(Jun activation domain binding protein-1,JAB-1)和S期激酶相关蛋白-2(S-phase kinase-associated protein-2,SKP-2)在瘢痕疙瘩、增生性瘢痕组织中的表达及其与肿瘤抑制因子P27^kip1之间的相互关系,探讨它们在病理性瘢痕形成中的作用及机制。方法应用免疫组织化学链霉亲和素-过氧化物酶(SP)染色法结合计算机病理图像分析,检测瘢痕疙瘩、增生性瘢痕及正常皮肤组织中的JAB-1、SKP-2及肿瘤抑制因子P27^kip1蛋白的表达水平,并进行统计学分析。结果瘢痕疙瘩、增生性瘢痕中JAB-1和SKP-2的表达均较正常皮肤高,而P27^kip1的表达则较正常皮肤低;瘢痕疙瘩与增生性瘢痕之间的各表达指标间比较无明显差异。病理性瘢痕组织中JAB-1和SKP-2的表达均与P27^kip1蛋白表达呈负相关性(分别为r=-0.630,P〈0.01和r=-0.538,P〈0.01)。结论JAB-1和SKP-2在瘢痕疙瘩、增生性瘢痕组织中的表达异常增高,促使细胞周期负性调控因子P27^kip1蛋白的降解,从而影响创伤的愈合过程,在瘢痕疙瘩、增生性瘢痕形成中起重要作用。Objective To study the expression of JAB land SKP-2 in the pathological scars and their relationship with P27^kip1 level, and to investigate their roles and probable mechanisms in the patho genesis of abnormal scars. Methods Immunohistochernical technique (SP) was performed to detect the expression and distribution of JAB-1, SKP 2 and P27^kip1 in keloid, hypertrophic scar and normal skin. Statistics was used to analyze the data. Results Both JAB-1 and SKP-2 expressed much higher in abnormal scars than in normal skin. P27^kip1 level in normal skin was much higher than in abnormal scar. There was a significant inverse correlation between the expression of P27^kip1 and JAB1 (r =-0. 630, P〈0.01). And there was a significant inverse correlation between the expression of P27^kip1 and SKP-2 (r=-0.538,P〈0. 01). There was no significant difference between keloid and hypertrophic scar in JAB-1, SKP-2 and P27^kip1, Conclusions The results indicate that the expression of JAB-land SKP-2 is remarkably increased, both of which may be contribute to the low expression level of the P27^kip1 in abnormal scar and be possible factors for keloid and hypertrophic scar formation.
