摘  要:目的 观察睫状神经营养因子(CNTF)对培养的大鼠视网膜神经节细胞(RGCs)bcl-2表达的影响。方法 取出生后2~3d Wistar大鼠视网膜组织行RGCs培养,Thy-1单克隆抗体免疫细胞化学鉴定RGCs。实验分2组,对照组单纯加入DMEM,CNTF组加入浓度为20ng/ml的CNTF。RGCs培养3d后CNTF组和对照组分别行Westernblot检测RGCs内bcl-2蛋白的表达。结果 Thy-1单克隆抗体免疫细胞化学检查显示,培养3d的RGCs胞膜和轴突呈棕黄色,且90%以上为免疫阳性细胞。CNTF组RGCs内bcl-2蛋白表达高于对照组(P〈0.05)。结论 CNTF对神经节细胞的营养作用可能与bcl-2基因表达增高有关。Objective To study the effect of ciliary neurotrophic factor (CNTF)on expression of bcl-2 in cultured rat retinal ganglion cells. Methods The retinal ganglion cells (RGCs) were cultured with the retina of Wistar rats 2-3 postnatal days. The RGCs cultured for three days were identified by Thy- 1.1 monoclonal antibody immunohistochemistry. There were two groups, ie, control group ( added with DMEM) and CNTF group ( added with 20 ng/mL-1CNTF). The expression of bcl-2 protein in RGCs that cultured for three days in CNTF group and control group was determined by Western blot. Results The cell membrane and neurite of RGCs cultured for three days were dyed huffy by Thy-1.1 antibody with immunohistochemistry method and the positive rate of retinal ganglion cells was over 90%. Western blot analysis showed that the expression of bcl-2 protein in CNTF group was higher than that in control group, with statistical difference ( P 〈 0.05 ). Conclusions CNTF can increase bcl-2 expression of cultured RGCs in vitro, which suggests that the nutrien effects of CNTF in retinal ganglion cells may relate to the increased expression of bcl-2 protein.
