摘 要:目的 探讨大鼠严重烧伤后,在脑水肿形成过程中水通道蛋白-4(aquaporin-4,AQP4)的表达变化。方法 采用30%TBSAm度烧伤大鼠模型,用干湿重法和免疫组织化学方法分别检测烧伤后不同时间段脑含水量和AQP4的表达变化。结果 严重烧伤后2h脑含水量和AQP4表达增加;烧伤后6h达到高峰,烧伤后48h仍高于正常;AQP4的表达与脑含水量呈显著正相关(r=0.8703,P〈0.01)。结论 严重烧伤后AQP4表达明显增强,提示AQP4与烧伤后脑水肿的形成密切相关。Objective To study the expression changes of aquaporin-4 (AQP4) protein in cerebral edema after severe burn. Methods Rats inflicted with 30% TBSA IU burn were taken as the model. The changes of cerebral water content (CWC) were detected by the dry-wet weight methods. Immunohistochemistry was used to study the expression change of AQP4 at different times after severe burn. Results Expressions of CWC and AQP4 were increased two hours postburn, reached its peak at the 6th hour and still higher than normal level 48 hours after severe burn. The expression of AQP4 was correlated with CWC after severe burn ( r = 0. 8703, P 〈 0. 01 ). Conclusion Expression of AQP4 increases after severe burn, as indicates that AQP4 has a correlation with the formation of cerebral edema after severe burn.
