摘  要:目的:探讨肉苁蓉总苷(GCs)对双侧颈总动脉结扎大鼠学习记忆功能的影响及其机制。方法:雄性SD大鼠,行双侧颈总动脉结扎术,以银杏叶片为阳性对照,术后2 wk,进行大鼠跳台试验和Y-型迷宫试验,观察GCs对大鼠学习记忆障碍的保护作用;并测定GCs对大鼠脑组织中超氧化物歧化酶(SOD)、谷胱甘肽-过氧化酶(GSH-Px)活性、丙二醛(MDA)含量和乙酰胆碱酯酶(AchE)活性的影响;采用NBT染色法测定24h后脑梗死范围;行为指标采用11分制进行评分;TUNEL法检测脑细胞凋亡。结果:GCs能提高双侧颈总动脉结扎所致的脑缺血大鼠学习记忆水平;提高脑组织SOD、GSH-Px活性,降低MDA含量及AchE活性;改善神经症状;降低脑细胞凋亡率;缩小脑梗死范围。结论:GCs对双侧颈总动脉结扎大鼠认知功能障碍有防治作用,其作用机制可能与增强自由基清除酶活性,防止脂质过氧化反应,抑制细胞凋亡有关。还可能与通过抑制AchE活性,维持脑内乙酰胆碱(Ach)的正常水平有关。AIM: To evaluate the effects on leaning-memory function of glycosides of cistanche (GCs) of rat model undertaken bilateral common carotid artery ligation and its possible mechanism. METHODS: Male Sprague-Dawley rats undertook bilateral common carotid artery ligation and Yinxingye tablet was used as the positive control medicine. Two weeks after the ligation, the effects of GCs on learning and memory were observed by using step-through test and step-down test and Y-maze test. The activities of SOD, MDA, GSH-Px and AchE in cerebrum were simultaneously recorded. The infarct area of brain was measured by nitrobenzene thiocyanate (NBT) staining technique 24 h later. Neurological deficits were evaluated by 0-11 scales and cerebral cellular apoptosis by TUNEL test. RESULTS: GCs improved learning-memory deficiency in rat undertaken bilateral common carotid artery ligation; increased the activities of SOD, GSH-Px in rat cerebrum; reduced the activities of AchE and contents of MDA and infarct area in rat cerebrum; improved neurological symptoms and decreased the rate of cellular apoptosis. CONCLUSION: GCs can improve the leaning-memory ability of rat undertaken bilateral common carotid artery ligation, through possible mechanism of enhancing free radicals and clearance of enzymatic activities, inhibition of lipid hyperoxidation, maintain the normal level of Ach, and inhibition of cerebral cellular apoptosis.
