摘 要:目的:建立癫痫患儿丙戊酸血药浓度的测定方法,探讨其血药浓度与剂量、患儿性别、年龄及临床疗效的相关性。方法:采用高效液相色谱法测定122例癫痫患儿单一服用丙戊酸达稳态后(10~14d)的血药浓度,并分析其结果。结果:丙戊酸血药浓度在10~180μg·mL-1范围内呈良好线性关系(r=0·9989);不同年龄患儿丙戊酸血药浓度与剂量有一定差异,而血药浓度与患儿年龄、性别无显著相关性;血药浓度在40~100μg·mL-1范围内临床疗效好。结论:本方法简便、准确、专一性强,可用于癫痫患儿丙戊酸血药浓度监测,并为临床用药提供参考。OBJECTIVE:To establish a RP- HPLC method for the determination of serum concentration of valproic acid in epileptic children and to study the correlation between serum drug concentration with dosage, patients' sex, age and clinical efficacy.METHODS: The serum concentration of valproic acid in 122 epileptic children after oral administration of single valproic acid(10-14d) was determined by HPLC and the determination results were subjected to analysis.RESULTS: The linear range for valproic acid was 10-180μg·mL^-1 (r = 0.998 9) .There were a little difference between serum concentration and the dosage of valproic acid in epileptic children with different age, but serum drug concentration showed no significant correlation with patients' age and sex difference.The optimal therapeutic concentration window of valproic acid was 40-100μg·mL^-1.CONCLUSION: The established method is simple, accurate and specific.It is applicable for the monitoring of serum concentration of valproic acid and it can provide reference for rational use of drugs in the clinic.
