摘 要:目的:体外比较不同浓度硫贲妥钠、咪唑安定对成年人Th0细胞分化的影响。方法:采集20例健康志愿者外周静脉血,进行全血和外周血单个核细胞(PBMCs)培养。每一份血样均用于下列各组研究:①空白对照组(C组);②不同浓度硫贲妥钠组[4μg/ml(T1组)、20μg/ml(T2组)和40μg/ml(T3组)];③不同浓度咪唑安定组[50ng/ml(M1组)、150ng/ml(M2组)和500ng/ml(M3组)]。培养的全血和PBMCs均用相应浓度药物预先作用24h,然后加入相应的刺激剂。刺激后检测胞内细胞因子(IFN-γ+/IL-4+)和细胞膜表面趋化因子受体(CCR5+/CCR3+)含量评估Th0细胞的分化情况。结果:分别与C组和T1组相比较,T2、T3组的Th1细胞亚群比例均明显降低(P<0.05);与T1组相比较,T3组的Th2细胞亚群比例明显降低(P<0.05)。咪唑安定各组Th1、Th2细胞亚群的比例无显著改变。结论:硫贲妥钠对Th1细胞分化有明显抑制作用,使Th0细胞向Th2方向分化;咪唑安定对Th0细胞分化无明显影响。Objective:To observe the effect of different concentrations of thiopental and Midazolam on the differentiation of T helper cells in vitro in adults. Methods:Peripheral venous blood samples were collected from 20 middle-aged healthy donors. In each volunteer,20 rnl of blood was divided into 7 groups:control group,3 thiopental groups(4 μg/ml,20 μg/ml, 40 μg/ml),3 midazolam groups(50 ng/ml, 150 ng/ml, 500 ng/ml). Whole blood and mononuclear cells from peripheral blood(PBMCs) were incubated with different concentrations of thiopental and midazolam for 24 h, while the control group without any drugs. Then the stimulants- phorbolmyristate+lonomycin+glogistop were added to whole blood and phytohemagglutinin(PHA) was add to PBMCs. The whole blood was incubated for another 4 h and PBMCs were incubated for another 48 h. Then the T-lymphocytes were collected for determination of intracellular level of IFN-γ(as a marker of Thl cells) and IL-4(as a marker of Th2 cells) in the whole blood using flow cytometry and the expression of CCR5+(as a marker of Thl cells) and CCR3+(as a marker of Th2 cells) in PBMCs. Results:Compared with the control group, in thiopental 20 μg/ml, 40 μg/ml group, the percentage of Thl ceils decreased significantly. Among different concentration in thiopental groups, the percentage of Thl cells in high-dose or middle-dose group was obviously lower than that in low-dose group, the percentage of Th2 cells in high-dose was obviously lower than that in low-dose group. In midazolam group, there was no significant change. Conclusion :Thiopental can inhibit Th 1 activity and shift the balance towards immunosuppressive Th2-1ike responses predominantly. Midazolam has no significant effect on Thl and Th2 activity.
