摘 要:采用抽吸法、切剖法、刺破法采集山羊卵母细胞。抽吸法得到的卵母细胞数最多,但花费时间最多且裸卵较多,不适合回收卵母细胞。切剖法与刺破法差异不显著,但切剖法所用时间多于刺破法,两者差异显著(P<0.05)。添加20、30、50 ng/ml表皮生长因子(EGF)于卵母细胞培养液中与对照组相比,20 ng/ml EGF对卵母细胞的成熟没有显著影响;30 ng/ml EGF可显著促进卵母细胞的成熟(P<0.05),50 ng/ml EGF可极显著促进卵母细胞的成熟(P<0.01)。Follicular oocytes were recovered using three different methods (aspiration, dissection and puncturing). The average total number of oocytes recovered per ovary was significantly higher by the aspiration method. But the time required for processing each ovary and usable oocytes by the aspiration were comparatively more, so aspiration was not a good method to recover oocytes. The numbers of oocytes recovered per ovary were 5.6 and 6.7 for dissection and puncturing, respectively. The percentages of oocytes of A, B and C grade for dissection were 49 %, 43 % and 8 %, and for aspiration were 52 %, 42 % and 6 %, respectively. There were no significant differences between the two methods (P〈0.05). But time required by dissection was more than puncturing. Growth culture medium was supplemented with EGF of 20 ng/ml, 30 ng/ml and 50 ng/ml. Compared with control group, EGF (30 ng/ml) significantly promoted maturation of the oocytes, but had no stimulatory effect on cleavage of the zygotes. EGF (50 ng/ml) extremely significantly promoted the maturation of the oocytes and the cleavage of the zygotes.
