摘　　要：目的:探讨人骨髓间充质干细胞(hMSCs)体外分离培养及鉴定。方法:成人骨髓取自手术中非血液系统疾病、非肿瘤及乙肝表面抗原阴性患者摘取的肋骨,采用贴壁筛选法进行体外扩增;用免疫组织化学法对培养的细胞进行鉴定。结果:体外培养的原代hMSCs9d融合,免疫组织化学染色CD44表达阳性。结论:贴壁筛选法在体外很容易分离培养和扩增hMSCs,可作为获得hMSCs的一种有效手段。Objective： To investigate the method of isolation, culture, and identification of human mesenchymal stem cells （hMSCs）. Methods： Human bone marrow was obtained, and hMSCs were isolated and purified by using direct anchoring and identified by using immunohistochemical method, Results： Primary cultured hMSCs reached confluence within 9 days, hMSCs were positive for CD44, Conclusion： hMSCs can be isolated, cultured, and expanded easily by using direct anchoring, which can be used as an effective method for culturing hMSCs.
