摘 要:目的:探讨慢性铝暴露损害大鼠学习记忆的作用机制。方法:选择断乳后Wistar大鼠,以含不同浓度氯化铝的水进行饲养。3个月后,进行跳台行为学及有丝分裂原活化蛋白激酶(MAPK)活性的测定。结果:染铝各组大鼠的血铝、脑铝浓度与对照组相比均升高,且有显著性差异(P<0.01);染铝各组潜伏期与对照组比较均缩短(P<0.01);染铝组的海马MAPK活性与对照组比较显著降低(P<0.05)。结论:不同浓度慢性铝暴露可引起大鼠脑铝和血铝的含量增高,使海马MAPK活性下降,导致大鼠认知功能降低。Objective: To explore the mechanism of the impairments of longterm potentiation (LTP) and learning and memory function induced by chronic aluminum exposure by detecting the activity of mitogen-activated protein kinase (MAPK) in rats. Methods: The ablactated Wistar rats were fed with different concentrations of AlCl3 in the drinking water. The cognitive ability and and the activity of MAPK in rat hippocampus were determined by diving tower method and modified Takai method 3 months later, respectively. Results:The levels of aluminum in the blood and brains of rats exposed to different concentrations of AlCl3 were significantly higher than those in control subjects ( P 〈 0.01 ). Compared with control subjects, the latent period was significantly shorter and the activity of MAPK significantly lower in rats exposed to different concentrations of AlCl3 ( P 〈 0.01, P 〈 0.05 ). Conclusion: Chronic aluminum exposure can not only increase the concentration of aluminum in blood and brains but also reduce the activity of MAPK, so it may impair the cognitive ability in rats.
