摘 要:目的:探索骨髓基质细胞体外定向诱导分化为神经细胞的可能性。方法:取大鼠的骨髓基质细胞在体外培养,用免疫组化方法检测正常脊髓提取液和脑源性神经生长因子诱导分化,观察诱导后细胞的形态。结果:骨髓基质细胞在体外培养呈梭形。诱导后细胞呈神经元样细胞改变。神经元烯醇化酶和胶质纤维酸性蛋白染色阳性。结论:骨髓基质细胞可在体外诱导分化。Objective: To investigate the possibility of induced differentiation of rat bone marrow stromal cells (BMSCs) into neural cells in vitro. Methods: BMSCs were collected from rat femur and cultured in a medium containing the extracts of normal spinal cord and brain-derived neurotrophic factor. The morphological changes of the cells were observed. The cultured cells were stained immunocytochemically with neuron-specific enolase (NSE) and glial fibriliary icidic protein (GFAP) antibodies. Results: BMSCs were successfully cultured in vitro, The induced cells became neuron-like cells and were stained positively for NSE and GFAP antibodies. Conclusion: BMSCs can differentiate into neuron-like cells in vitro.
