胎盘组织KiSS-1 mRNA及其蛋白肽metastin表达在子癎前期发病中的意义

摘　　要：目的:探讨胎盘组织中KiSS-1mRNA及其蛋白肽metastin表达在早发重度子癎前期发病中的作用。方法:采用RT-PCR、Western blot、免疫组化方法检测57例子癎前期患者(轻度15例和重度42例,其中早发重度12例,晚发重度30例)和30例正常晚期妊娠者胎盘组织中KiSS-1mRNA及其蛋白肽metastin的表达水平和定位。结果:胎盘组织中蛋白肽metastin主要表达在绒毛的合体滋养细胞和细胞滋养细胞。子癎前期组胎盘组织中KiSS-1mRNA及蛋白肽metastin表达分别为(1.73±0.24)和(78.41±7.96)μg/100μg总蛋白,显著高于正常晚期妊娠组(P均<0.01),以早发重度子癎前期组尤为显著。结论:子癎前期存在KiSS-1mRNA及其蛋白肽metastin表达增强,且早发重度子癎前期组表达显著增高,可能是早发重度子癎前期发病的主要原因之一。Objective： To investigate the expressions of KiSS-1 mRNA and metastin, its peptin, in the placentas of women with early-onset severe preeclampsia and their role in the pathogenesis of early-onset severe preeclampsia. Methods： The expressions and cellular distributions of KiSS -1 mRNA and metastin were determined by reverse transcription polymerase chain reaction, Western blotting, and immunohistochemical method in the palcentas of 57 women with preeclampsia, including 15 with mild preeclampsia and 42 with severe preeclampsia （ 12 with early-onset severe preeclamsia and 30 with late-onset severe preeclamsia） , and 30 normal women in late pregnancy. Results： Metastin were mainly located in the syncytiotrophoblasts and cytotrophoblasts. The expression levels of KiSS -1 mRNA and metastin in women with preeclampsia were 1.73 ±0.24 and 78.41 ±7.96 μg per 100 μg total protein, both of which were significantly higher than those in normal women in late pregnancy （ P 〈 0.01 and P 〈 0.01 ） , especially in women with early-onset severe preeclampsia. Conclusion：The expressions of KiSS -1 mRNA and metastin increase in women with early-onset preeclampsia, especially in women with early-onset severe preeclampsia,which may be involved in the pathogenesis of early-onset severe preeclampsia.
