摘 要:在细胞中导入dsRNA会引起体内同源基因特异性的沉默(silencing),这种现象被称为RNAi(RNA interference).该技术经过十年的发展,逐步走向成熟,并引起了生物科学的一次新的革命。它被广泛应用于细胞信号转导、发育生物学、基因功能学及肿瘤的基因治疗等多个领域,并取得了一系列开创性的成果。肿瘤细胞对抗癌药物产生抗药性是化疗失败的一个主要原因。而多药耐药现象是系指肿瘤细胞对一种抗肿瘤药物出现耐药性的同时,对其他多种结构不同、作用靶位不同的抗肿瘤药物也有耐药性的现象。利用RNAi技术研究和治疗肿瘤的多药耐药性为困扰医学界二十多年的难题找到了一个新的突破口。the guide—in of dsRNA to cells will lead to the silence of the specialty of homology genes, the phenomenon is called RNAi.This technique has come to mature through 10 years development,and it arises a revolution in life science.It has been widely used in many fields including the transmission of cell signal,growth biology,gene function and gene treatment to tumor,etc,and it has achieved a series of creative results.The drug resistance of tumor cell to anti—cancer medicine is one of the failures of chemotherapy.While the phenomenon of MDR refers to MDR of tumor cell has drug resistance to anti—tumor drug at the same time to anti—cancer drugs of different structure and different target.With RNAi technology research and therapy to the MDR of tumor have made a new breakthrough for the puzzlement of medical field for 20 years.
