摘 要:从番茄基因组中克隆LeNCED1基因5′-侧翼1456 bp调控序列,经生物信息学分析表明,LeNCED1基因上游调控序列中存在响应昼夜节律的顺式作用元件Circadian、光响应元件Box I、逆境胁迫响应元件(TC-rich repeats)、ABA响应元件ABRE等,构建了LeNCED1基因上游调控序列与报告基因GUS融合的植物双元表达载体pLeNCED1p:GUS。The cloning of 1456-bp 5′-flanking sequence of LeNCED 1 gene from tomato and its fusion with β-glucuronidase gene (GUS) were reported. The flanking sequence was bioinformatically analyzed in the database of plant cis-acting regulatory element (PlantCARE). The result showed that there were several important cis-acting elements, including Circadian (response to circadian rhythm), Box I (response to light), TC-rich repeats (response to stress) and ABRE (response to ABA), in the upstream regulatory region of LeNCED1 gene. The binary vector harboring 5′-flanking sequence of LeNCED1 gene fused with GUS gene originated from pCAMBIA1301 was constructed to be used in coming plant transgene.
