摘  要:目的应用高效液相色谱法测定栽培射干中次野鸢尾黄素的含量,考察其药材的内在质量。方法采用高效液相色谱法,色谱柱为TSK ge1 ODS 80 TS250 mm×4.6 mm;流动相为甲醇-磷酸水溶液(pH=3)∶53%∶47%;流速为1 ml/min;检测波长为266 nm;柱温35℃。结果次野鸢尾黄素在0.0118-0.118μg范围内与色谱峰面积呈线性关系(r=0.9999);平均加样回收率为99.84%,RSD为2.059%,次野鸢尾黄素的含量为0.1252%。结论该法简便、准确、灵敏度高,栽培射干中的次野鸢尾黄素的含量大于0.10%,符合《中国药典》要求。Objective To determine irisflorentin in cultivated Belarncanda chinensis( L. )DC by HPLC, and to inspect the quality of its ingredients. Methods Using HPLC with TSK gel ODS Column80(250 mm ×4.6 mm) ,the mobile phase was composed of a mixture of methanol - phosphoric aicd aqueous soluton ( pH = 3 ) : 53% : 47%, the flow rate was 1 ml/min, the UV detection wavelength was set at 265 nm,and the temperature was at 35℃. Results Irisflorentin showed linear relationship with peak areas in the range of 0.0118 -0.118 μg( r = 0. 999 9 ). The average recovery was 99.84% , RSD was 2.509%. Conclusion The method is simple,rapid and accurate. The quantity of irisflorentin in Cultivated Belamcanda Chinensis (L.)DC exceeds 0.10% and meets the Chinese Pharmacopoeia.
