摘　　要：目的:研究转化生长因子β1(TGF-β1)对同种反应性T细胞增殖能力及CD25表达水平的影响。方法:以丝裂霉素处理的Balb/C(H-2d)小鼠脾细胞为刺激细胞,羧基荧光素乙酰乙酸(CFDA-SE)标记的C57BL/6(H-2b)小鼠T细胞为反应细胞进行混合淋巴细胞培养(MLC)。分别设立对照组(TGF-β10 ng/ml)、低浓度组(TGF-β11ng/ml)、中浓度组(TGF-β15 ng/ml)和高浓度组(TGF-β120 ng/ml)。MLC结束后利用Alamar Blue检测T细胞增殖活性,流式细胞仪检测CD3、CD25表达水平。同时设立白细胞介素-2(IL-2)组(TGF-β15ng/ml+IL-2 100U/ml),以观测其对TGF-β1生物学效应的影响。结果:TGF-β1可明显降低同种抗原活化T细胞的增殖反应强度,而且随着TGF-β1剂量加大,免疫抑制能力增强。TGF-β1在抑制T细胞增殖的同时,也可使CD25+T细胞比例上调,但并无明显的剂量依赖性。加入外源性IL-2后,T细胞增殖活性增强,同时CD25+T细胞比例降低。结论:TGF-β1可明显抑制同种抗原活化T细胞的增殖,并使CD25+T细胞比例增加,而外源性IL-2则可逆转TGF-β1的免疫抑制功能,下调CD25+T细胞比例,说明外源性IL-2可以影响TGF-β1的生物学效应。Objective ：To investigate the roles of TGF-β1 in the proliferation and CD25 expression of alloreactive T cells. Methods：In MLC, purified C57BL/6 T cells labeled with CFDA-SE were added to the Balb/c splenocytes pretreated with mitomycin. Five groups were established： control group（TGF-β1 0 ng/ml）, low concentration group（ TGF-β1 1 ng/ml）, middle concentration group（ TGF-β1 5 ng/ml）, high concentration group（ TGF-β1 20 ng/ml） and IL-2 group（ TGF-β1 5ng/ml ＋ exogenous IL-2 100 U/ml）. After MLC completed, the proliferation of T ceils was tested by Alamar blue method, and the expression of CD3 and CD25 was detected by flow cytometry. Results：TGF-β1 suppressed the proliferation of T ceils activated by alloantigen and the suppression could be enhanced as the TGF-β1 concentration increased. At the same time, TGF-β1 increased the ratio of CD25 ^＋ T ceils with dosage independence. In addition, the exogenous IL-2 could strengthen the proliferation of alloreactive T ceils , and decreased the ratio of CD25^＋ T cells（P 〈 0.05 ）. Conclusion ： TGF-β1 suppressed the proliferation of alloreactive T ceils, and increased the ratio of CD25^ ＋T ceils, the biological effect of TGF-β1 could be reversed by after adding the exogenous IL-2.
