摘 要:目的观察乙酰肝素酶(HPSE)在酒精诱导的大鼠肝纤维化时的动态表达,以探讨其在肝纤维化形成过程中的作用。方法采用酒精-玉米油-吡唑灌胃诱导肝纤维化模型,分别用免疫组织化学染色.和原位杂交方法检测乙酰肝素酶的蛋白和mRNA在大鼠酒精性肝纤维化发生发展中的动态表达。HE、Masson染色判断肝纤维化程度。结果乙酰肝素酶在酒精性肝纤维化组表达阳性率明显高于正常肝组织(8/17vs0/11,P〈0.005),有显著的统计学差异。乙酰肝素酶的蛋白、mRNA表达模式和定位基本一致。结论乙酰肝素酶参与了早期肝纤维化的形成和发展,且乙酰肝素酶的合成与储存在同一细胞。Objective To explore the significance of heparanase in rat hepatic fibrosis induced by alcohol. Methods In rat models of alcoholic fibrosis, the expression of heparanase mRNA and protein were detected by in situ hybridization and by immunohistochistochemistry. The tissue specimens were stained with HE and Masson methods to evaluate the grade of liver fibrosis. Result The expression of heparanase mRNA and protein were up-regulated in the alcoholic hepatic fibrosis and higher than that of normal liver tissues (8/17 vs 0/11, P〈0.005). Moreover, a similar pattern of expression was observed both at mRNA and protein level. Conclusions It can be concluded that heparanase is involved in the pathogenesis of liver fibrosis. In addition, the co-expression of heparanase mRNA and protein implies that heparanase is synthesized and stored in the same ceil.
