高效液相色谱法测定首乌散中大黄素含量

摘　　要：建立了首乌散中大黄素含量的HPLC测定方法。采用Waters C18色谱柱（10μm，250mm×4．6mm），检测波长254nm，流动相为甲醇-水-磷酸（850：150：1），流速1mL/min。结果显示，大黄素在1．0～40．0μg／mL范围内，含量与峰面积呈良好的线性关系，相关系数为0．9999（n=8），平均回收率95．5％，RSD为2．0％。本方法简便、重现性好，结果准确可靠，可有效地控制首乌散的质量。A high performance liquid chromatographic method was reported for the quantitative determination of emodin in Shouwu San with Waters spherical Cls column（250mm × 4.6mm, 10μm）. The detecting wavelength was 254 nm;mobile phase was methyl alcohol-water-phosphor acid（850： 150：1 ） ;flow rate was 1.0 mL/min. The liner relationship ranged from 1.0 to 40 μg/mL with a correlation coefficient of 0.999 9. The average recovery was 95.5% with RSD of 2.0%. The method is simple, sensitive and accurate. It can be used in the quality control of Shouwu San.
