摘  要:目的:观察补肾益骨方对去势大鼠成骨细胞增殖及骨形态发生蛋白-2(bone morphogenetic protein-2,BMP-2)表达的影响。方法:取12周龄Wistar雌性大鼠40只,随机分为A(模型组)、B(治疗组)、C(阳性对照组)、D(正常对照组)四组,每组10只,前3组行完整双侧卵巢摘除术,D组行假手术,术后常规饲养12周,再分别予以生理盐水、补肾益骨方水提液、α-D3水溶液及生理盐水灌胃治疗12周,处死大鼠,取右胫骨上端做病理切片,通过HE染色观察成骨细胞增殖、免疫组化染色观察BMP-2表达。结果:治疗组成骨细胞数量明显增加,而且通过免疫组化染色发现BMP-2棕色深染的阳性细胞比例明显高于模型组。结论:补肾益骨方能促进成骨细胞分化、增殖,加速骨形成,具有治疗骨质疏松的作用。推测骨中BMP-2含量增加是其重要的治疗机理之一。Objective:To study the efficacy of Bushenyigufang on BMP-2 expression and osteoblast proliferation in ovariectomized(OVX) rats. Methods:Female Wistar 40 rats were divided into 4 groups:model, therapy, positive contrll and sham operation group(group A,B,C,D). Ovaries of group A,B,C were cut off,the other did not derl anything through laparotomy. Saline,bushenyigufang extractant,α-D3 aqueous solution, saline were given respectively during 12 weeks after operation. All rats were killed in week 12 after administration. The proximal end of the right tibia were cut for the pathological section,and osteoblasts were observed by HE staining and BMP-2 expression in bone tissue by immunohistochemistry staining. Results:Compared with group A, the expression of BMP--2 and the number of osteoblasts were significantly in- creased in group B. Conclusions: Bushenyigufang obviously increased the expression of BMP--2 and the number of osteoblasts in OVX rats and prevent bone from lossing effectively. The results showed that the high expression of BMP--2 may be an important therapeutic mechanism in osteoporosis.
