摘 要:取广西牛源大片吸虫成虫制备出分泌排泄(ES)抗原,用SDS-PAGE电泳,考马斯亮蓝染色显出7条蛋白带。免疫印迹试验(EITB)测出其主要特异性免疫成分分子量为36.3,28.5及25.5kDa,其中28.5与25.5kDa与法国肝片吸虫ES的部分抗原成分相同。两种片形吸虫与抗血清可以交叉使用无特异性,但与日本血吸虫抗原及抗血清均无交叉反应,人工感染大片吸虫2周后的兔血清即能识别此抗原。ES抗原提取方法简便,特异性好,敏感性高,可用于片形吸虫病免疫诊断。Fasciola gigantica excretory secreting(Fg. ES) products were prepared by using adult worms from native cattle. At least 7 bands of M r. Ranging from 25.5 to 44 kD a were indicated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE). Three peptides weighing 36.3,28.5 and 25.5 kD a were obtained by reacting with antiserum to Fasciola hepatica ES (Fh.ES) products in the enzyme linked immunoelectrotransfer blot assay (EITB), of which 2 peptides of 28.5 and 25.5 kD a with the same components as that of F.hepatia ES from France.Dot ElISA showed that the two ES products reacted with the antisera to Fh.ES and Fg.ES crossively,but no cross reactivity with Schistosoma japonicum worm antigen(SjwA) and antiserum.Fg.ES products could be recognized by the antiserum from infected rabbits as early as 2weeks post exposure with F.gigantica. However,it suggested that Fg.ES products have the highest potential as immunodiagnosis antigens for fasciolasis.
