鸡马立克氏病毒Ⅰ型毒株的分离和鉴定

摘　　要：从鸡马立克氏病（ＭＤ）疫苗免疫的发病鸡群中采集肝、脾、血液等病料，经鸭胚成纤维细胞（ＤＥＦ）培养盲传数代，分离到１株（ＭＤＶ）暂定名为沪Ｘ株。该病毒在ＤＥＦ培养上出现典型的ＭＤＶ细胞病变。电镜下病毒大小为１００ｎｍ左右，呈椭圆半开圆形，位于细胞核内。细胞培养物经饱和硫酸铵浓缩后，与标准的马立克氏阳性血清出现琼扩沉淀线。病毒按种ＳＰＦ鸡后，鸡群主要表现内脏肿瘤，并回收到ＭＤＶ，表明分离的沪Ｘ株具有致病性。A virus strain temporarily named as Hu X was obtained from the liver, spleen and blood ofMarek’s disease(MD) vaccinated chicken flocks showing clinical symptoms after several blind passages in duck embryo fibroblast(DEF).The isolated rirus appeared to be cytopathic effect(CPE) characterized by clusters of rounded,refractile degenerating cells in DEF .Electronic microscope showed that the isolated virus was an icosahedral virus with approximately 100nm in diameter.The isolated virus reacted with an antiserum against MDV in agar gel precipitation tests. Pathogenicity of the isolated virus was confirmed by inoculation of the isolated virus into l day old specific pathogen free chicks and demonstration of visceral tumors.The virus was also recovered from experimentally infected chickens.From the data shown abown above,the isolated virus was identified as a MDV stereotype Ⅰ.
