摘 要:在添加浓度为100mg/L秋水仙素的继代培养基上,对体积约为0.5cm3和0.3cm3的当归愈伤组织分别处理10d、20d、26d和35d,并在处理结束后的第1d和第10d取样,观察细胞倍性的变化趋势。结果表明:处理时间对4x细胞比例有极显著的影响;在处理结束后的恢复生长中,二倍体细胞明显增加,八倍体细胞显著减少,四倍体细胞则保持稳定;在长时间的诱导处理中。The changes of different ploidy cells in callus of Angelica sinensis sized about 0.5 cm 3 or 0.3 cm 3 and then treated by Colchicine with concentration of 100mg/L being added in the MS subculture medium for 10, 20, 26 and 35 days, respectively, were measured through examining the chromosome numbers in samples taken at 0 and 10 days after treatment. It was showed that durations of the treatment had significant effects on the percentages of 4x cells (31 26%~44 76%, P <0.01) and slight effects on 2x cells (23.91%~29.95%, P >0.05) and 8x cells (38.47%~44.38%, P >0.05) in the samples at 0 day. Therefore the changes of the relative percentages of these three kinds of cells in the callus subcultured for 10 days after the treatment were the 2x cells significantly increased ( P >0.01), 8x cells decreased ( P >0.01) and 4x cells remained at the similar levels compared to the observations at 0 day. The size of callus did not affect the chromosome doubling in the long term treatment by Colchicine.
