摘  要:目的探讨巯基乙酸(TGA)对卵母细胞体外成熟的影响。方法采用小鼠卵母细胞体外成熟和免疫荧光染色法,观察巯基乙酸处理后小鼠MⅠ/MⅡ期卵母细胞的成熟进程和形态结构。结果对照组、0.005mg/mlTGA组和0.020mg/mlTGA组生发泡破裂率分别为48.67%、62.67%和56.00%,第一极体排出率分别为26.47%、65.96%和45.24%,0.005mg/ml处理组第一极体排出率高于对照组和0.020mg/ml组,差异有统计学意义(P<0.05)。随着TGA处理剂量的增加,卵母细胞纺锤体和染色体形态结构异常的细胞比例和异常程度增加,0.020mg/mlTGA处理组与对照组有显著性差异。结论巯基乙酸可促进卵母细胞体外减数分裂的重启和成熟,但中期卵母细胞的纺锤体及染色体有结构缺陷,对后续胚胎发育可能会产生潜在影响。Objective To explore the effect of Thioglycolic acid (TGA) on maturating progress and structure of MⅠ/MⅡ stage oocytes in mice. Methods In vitro maturation and immunofluorescence stainings were adopted to observe the maturing progress and structure of MⅠ/MⅡ stage oocytes treated with TGA. Results The percentage of occytes with GVBD in control, 0. 005 mg/ml TGA and 0. 020 mg/ml TGA group were 48.67% were 62.67% and 56.00%, respectively, and the percentage of occytes with PB1 formation were 26.47%, 65.96% and 45.24%, respectively. The percentage of PB1 formation in 0. 005 mg/ml TGA group was significantly higher than that in control and 0. 020 mg/ml TGA group. Percentage and extent of abnormal configuration of spindle and chromosomes in oocytes at metaphase stage increased with the increasing of TGA dose. In contrast to the control, the percentage in 0. 020 mg/ml TGA group was significandy lower. Conclusions Improving resumption of meiosis and meiosis maturation of oocytes cultured in vitro and inducing aberration in configuration of spindle and chromosomes, TGA might have patent harm to subsequent embryo development.
