内皮素A受体拮抗剂BQ123对肉仔鸡股动脉压和肺动脉压的影响

摘　　要：为探讨内源性内皮素-1（ET-1）是否参与肉鸡血压的维持等生理调节活动，采用急慢性给予内皮素ETA受体的特异性拮抗剂BQ123，研究内源性ET-1对正常肉鸡股动脉压和肺动脉压的影响。结果表明：1）放免法检测2～6周龄正常肉仔鸡血浆（从（40．82±1.90）升高到（93．70±11．19）pg／mL）和肺匀浆内皮素（从（503．84±27．40）升高到（701．04±16．18）pg／g）的含量随日龄的增加有升高的趋势；2）急性翼静脉注射BQ123（0．4和2．0μg/kg）能够极显著降低肉鸡的股动脉收缩压、舒张压和肺动脉收缩压、舒张压（P〈0．01），注射2．0μg/kgBQ123压力下降程度比注射0．4μg／kg大，肺动脉压达到最低点的时间比股动脉早；3）慢性给予BQ123能够使肉鸡肺动脉收缩压极显著下降（对照组（3．37±0．47）kPa，BQ123组（2．46±0．50）kPa，P〈0．01），股动脉收缩压显著下降（对照组（20．95±1．17）kPa，BQ123组（18．85±2．05）kPa，P〈0．05）。试验结果显示：急性和慢性注射ETA受体的特异性拮抗剂BQ123都能降低肉鸡股动脉压和肺动脉压，推测内源性ET-1参与了肉鸡血压的维持等生理调节活动。Endothelin-1 （ET-1） is an important factor in the regulation of cardiovascular tone in humans and mammals, but its biological function in the avian vascular system has not been studied. The purpose of this study is to characterize the role of endogenous ET-1 in the vascular system of poultry by investigating the effect of endothelin [ a receptor （ETAR） antagonist BQ123] on femoral artery pressure （FAP） and pulmonary artery pressure （PAP） in broiler chickens. First, we found that plasma and lung homogenate ET-1 levels were both increased with age over seven weeks＇ life cycle of broiler chickens. Second, 60 min after intravenous injection of BQ123 （0.4 and 2.0μg/kg, respectively） there was a significant reduction in FAP and PAP （ P〈 0.05）. Third, continuous infusion of BQ123 （2.0μg/kg each time, two times a day） into abdominal cavities led to a significant decrease in the femoral artery systolic pressure （P 〈 0.05） and pulmonary artery systolic pressure （P〈 0.01） at 7 and 14 days after treatment. In summary, the ETAR antagonist BQ123 led to a significant reduction in FAP and PAP in broiler chickens, suggesting that endogenous ET-1 may be involved in the maintenance and regulation of systemic and pulmonary pressure in broiler chickens.
