曲狗草止血作用及初步安全性评价

摘　　要：目的探讨曲狗草氯仿-水相提取物的止血作用及初步安全性评价。方法通过家兔局部创面(皮肤、肝脏、股动脉)止血实验,评价曲狗草氯仿-水相提取物的止血作用;用LD50、皮肤刺激性实验和皮肤过敏试验,及动物的肝肾组织形态学,观察药物的安全性。结果该水相提取物的局部创面止血时间明显短于空白对照组(P<0.01)。LD50为483.08mg/kg。在一次性大剂量(300mg/kg)以下用药,病理组织形态学观察肝、肾等重要脏器未见异常变化;10%曲狗草氯仿-水相提取物,对实验动物完整皮肤及破损皮肤均无刺激性、过敏性及毒性反应。结论曲狗草氯仿-水相提取物具有良好的创面止血作用;该提取物毒性低,副作用小,用药安全可靠。Objective To investigate the hemostatic effects and safety of the QuGouCao chloroform-water extract. Methods The hemostatic property of the extract was studied by applying the extract soaked gauze to the local wound surface of skin, liver and femoral artery in rabbits. LDs0 test in Kuming mice was performed and the tissues of liver and kidney were collected for routine histochemical examination. The skin stimulus in rabbits and the sensitivity test in guinea pigs were conducted. Results The hemostatic time of wounds was obviously shorter （P 〈0.01 ） than that of blank group control. LD50 was 483.08 mg/kg. The liver and kidney of toxicated mice were normal when the extract less than 300 mg/kg. 10% QuGouCao chloroform-water extract had no stimulation effect on the intact skin or Conclusion The QuC, ouCao chloroform-water It is of low toxicity, few side effects and safety. damaged skin as well as caused no allergic or toxic reaction. extract has good hemostatic effect on the local wound surfaces.
