摘 要:目的:探讨复方苦豆子(CK)在逆转肾性高血压大鼠(RHR)左室肥厚的过程中,对心室肌Ca^(2+)含量及血浆血管紧张素Ⅱ(AngⅡ)水平的影响。方法:建立二肾一夹型高血压大鼠模型(2K1C-RHR),随机分为5组,即假手术组(n=10),模型组(n=11),CK高、中、低剂量组(剂量分别为33.60,16.80和8.40g·kg^(-1),n均为12)和卡托普利阳性对照组(8 mg·kg^(-1),n=10),另取10只作为假手术组。每日灌药一次,连续8周。每周测定各组动物尾动脉收缩压,在给药结束后,测定各组左室质量指数(LVMI)、左室壁厚度(LVWT)、心室肌Ca^(2+)及AngⅡ水平的影响。结果:用药1周后CK高、中剂量组血压显著下降(P<0.05),4周后降至最低水平(P<0.01)。CK高、中剂量组的LVMI,LVWT,心室肌Ca^(2+)含量及AngⅡ水平均显著低于模型组(P<0.01及P<0.05),CK高剂量与卡托普利作用相近。结论:CK对RHR的心脏重塑有一定的防治作用,其机制可能与钙拮抗作用及抑制肾素-血管紧张素系统(RAS)活性有关。Objective: To investigate the effects of compound kudouzi (CK) on left ventricular hypertrophy (LVH) , calcium in ventriclar myocardium and Ang Ⅱ in plasma of renal hypertensive rats. Methods: Two-kidney 1-clip (2K1C) renovascular hypertension was induced in male Wistar rats. Fifty- seven RHR were randomly divided into 5 groups as follows: model group( n = 11, fed with drinking water), high dose of CK group( CK-H group,n = 12,33.60 g·kg^-1·d^-1) , middle dose of CK group (CK- M group, n = 12,16.80 g·kg^-1·d^-1 ) , low dose of C K group ( C K-L group, n = 12,8.40 g·kg^-1·d^-1 ) and captopril group ( Cap group, n = 10,8 mg·kg^-1·d^-1 ) , in addition, sham-operated group ( sham group, n = 10)was fed with drinking water too. All rats were treated once a day for 8 weeks. The left ventricular mass index(LVMI) , the concentrations of calcium in ventriclar myocardium and angiotension Ⅱ (Ang Ⅱ )in plasma were measured. Results: Compared with model group, the LVMI were significantly decreased in CK-H and CK-M groups. The content of Ca^2+ in ventriclar myocardium of CK-H and CK-M groups was markedly decreased( P 〈 0.01, P 〈 0.05). The concent of Ang Ⅱ in plasma of CK-H group was much lower than that in model group[ (823.60 ± 211.24) vs( 1 345.54±+ 99.58) pg·mL^-1, P 〈 0. 001 ]. Conclusion:Chronic use of CK can inhibit and reverse ventricular hypertropy and the effect is dose-dependent. The mechanism may be related to its calcium antagonistic properties and the decreasing activity of RAS.
