摘  要:目的:探讨蕨麻(Potentilla anserina L.)对心肌细胞缺氧损伤的保护作用。方法:采用原代培养的SD乳鼠心肌细胞建立缺氧损伤实验模型,通过MTT法测定各组细胞代谢率,生化方法测定乳酸脱氢酶(LDH)和肌酸激酶(CK)的释放量以及细胞内超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果:蕨麻醇提物在24,12和6g·L^(-1)浓度能显著减少缺氧损伤心肌细胞LDH和CK的外漏量,并可显著提高细胞内SOD活性,减少MDA的产生。结论:蕨麻对心肌细胞缺氧损伤具有显著的保护作用,其机制之一可能是清除缺氧导致的心肌细胞自由基堆积,减少脂质过氧化。Objective:To study the protective effect of Potentilla anserina L. on neonatal rats cardiac myocytes injured by hypoxia. Methods: A model of hypoxia injury was established using primary cultural myocytes in neonatal SD rats . The metabolic ability of the cells was detected by MTT assay. The activities of lactate dehydrogenase(LDH) and creatine kinase (CK)out of cells and superoxide dismutase (SOD) in cells, and the productions of malondialdehyde (MDA) in cells were measured with biochemistry approaches. Results : 0. 024 g·mL^-1,0. 012 g· mL^ - 1 and 0. 006 g· mL^- 1 of the extract of Potentilla anserina L. in alcohol could significantly reduce the release of LDH, CK from myocytes and increase activity of SOD and inhibit productions of MDA in cells during hypoxia injury. Conclusion : Potentilla anserina L. has remarkably protective effect on hypoxia injury in neonatal rats myocytes.
