氨茶碱对A549细胞吞噬凋亡嗜酸粒细胞及分泌IL-6、IL-8的影响

摘　　要：目的探讨氨茶碱对A549细胞吞噬凋亡嗜酸粒细胞的作用及对白介素(IL)-6、IL-8分泌的影响。方法分离纯化人外周血嗜酸粒细胞,48h自然老化后进行凋亡鉴定,加入到不同浓度的氨茶碱干预后的A549细胞。动态观察A549细胞吞噬凋亡嗜酸粒细胞现象,并对已固定染色的细胞进行吞噬率统计。另将不同浓度的氨茶碱分别干预A549细胞、A549与凋亡的嗜酸粒细胞、A549与脂多糖,收获培养后的上清液,采用放射免疫法检测白细胞介素IL-6和IL-8的浓度。结果氨茶碱以时间和浓度依赖的方式抑制A549细胞吞噬凋亡的嗜酸粒细胞;氨茶碱抑制脂多糖刺激A549细胞后IL-6、IL-8的分泌且存在浓度依赖关系。结论氨茶碱抑制A549细胞吞噬凋亡的嗜酸粒细胞,可通过抑制IL-6、IL-8的方式发挥抗炎的作用。Objective To investigate the effects of A549 cells engulf apoptotic eosinophils and IL-6, IL-8 treated by aminophylline （AM）. Methods Eosinophils were purified from normal donors. The cells were cultured for 48 h, apoptosis induced and added into A549 cells which were treated by aminophylline. The phenomenon of A549 cells engulf apoptotic eosinophils was observed dynamically, and the ratio of engulfment calculateed was under the light microscope with the cells which were fixed and stained. Add aminophylline of different concentration to A549 cells, A549 cells and lipopolysaccharide（ LPS）, A549 cells and apoptotic eosinophils. All these supernatant liquid were collected and using radioimmunoassay kit to detect the concentration of IL-6, IL-8. Results Aminophylline inhibited the A549 cells engulf apoptotic eosinophils in time and concentration dependent manners ; Aminophylline inhibited. The changes of IL-6, IL-8. which induced by A549 cells excited with lipopolysaccharide. Conclusion Aminophylline can inhibit the A549 cells engulf apoptotic eosinophils, and the secretion of IL-6, IL-8 on anti-inflammation.
